A hard choice, but if I had to pick one to take with me to a desert island, I think it would be "Last Exit to Springfield". That episode just has it all: comedy, drama, weird dream sequences, and Mr Burns.

Or "Who Shot Mr Burns?", mostly because then I'd get two in one...
Pretty little baby
Pretty little monster
Went to the good school
Left with honours
Brand new tycoon

Not a question I can answer easily. But if I were to make a wild stab at it, I'd say 'Rosebud'. Every second of that episode was hilarious, even the scenes with no dialoge, like Mr. Burns' gate in the opening scene. (With the exception of Mr.Burns' younger brother George, in which I didn't get the joke. Before my time I guess)

Quotes:

Burns: [toward the Ramones] Have the Rolling Stones killed.
Smithers: Sir, those aren't --
Burns: Do as I say!

Homer: You know, Mr. Burns is so cheap --
Burns: What?
Homer: I mean, you know, Mr. Burns is so old --
Burns: How dare you!
Homer: Woo hoo, tough crowd.

Bart: Dad, I know you're discouraged, but please don't deny the world
your fat can.
Homer: Don't worry, boy, he'll be ready for your Aunt Selma's birthday.
Lisa: I knew it.

Homer: Maggie, I'm trying to watch TV.  Put that moldy old bear down![realizing] Moldy?  Old?
I'm gonna get something to eat!

Marge: I'm sure he'll offer a fair reward.  [as an afterthought] And then we'll make him double it.
Family: Huh?
Marge: [defensive] Well why can't I be greedy once in a while?

Marge: [incredulous] Have you been up all night eating cheese?
Homer: [slurred] I think I'm blind...

Barney: [brandishing a gun] Homer, give him what he wants!

{[A dumb sitcom is shown on TV]}
Burns: {Smithers, I'm home!  [canned laughter]}
Smithers: {What, already?  [canned chuckle]}
Burns: {Yes.  [loud canned laughter]}
Lisa: {[watching] Is it my imagination or is TV getting worse?}
Homer: {Ehh, it's about the same.  Uh oh!  Look out, Smithers!}
{[sound of breaking glass]}
{Heh heh, I love this show.}

So on and so forth...
Can you withstand the power of my super taunt?

That's a really hard choice indeed. It would be easier to make a top ten or at least a top three, but if I'd really had to choose one, I'd go with "Separate Vocations". It is the episode that changed me into a Simpsons fan, and esp. into a Lisa fan.

For me, it'd have to be Summer of 4 ft. 2.  With great moments too numerous to mention, this has always been my favourite (except for that brief moment when Trilogy of Error aired) and no doubt always will be.

Oh, what the heck.  I'll mention a few scenes.

BART: (signing yearbook) Your daughter's name is Seymour?
SKINNER: I...  Alright, I lied.  It's for me.

HOMER: (reading Flanders' letter) "Wel-diddly-elcome, Simpsons."  Oh, jeez, he actually wrote "diddly"!

LISA: (spooked by a seagull) It's not your fault... It's a bird thing, you don't control the birds, someday you will, but not now.

And of course, the dating game scene:

MARGE: (to Bart) You've got the Dud!
HOMER: (to Milhouse) Hey, he looks like you poindexter!
BART: How come Lisa gets to hang out with her friends?
HOMER: You got friends, you got the Dud right here! *elbows Milhouse* Stand up for yourself poindexter!

And perhaps my most favourite scene, the breakfast scene with Bart and Lisa.  I could put it down here, but it's really something you have to see and hear to fully appreciate. :)

I second that! Not only funny as hell, it introduced the coolest character on the show ever! :) (I said ever and I meant ever!)

Oh, for my favourite THOH segment, I agree with Rich. "Citizen Kang" is brilliant. (And it was brilliant even to a Swede who watched it at least a year after that election, too.)

Mine's one rather out of left field - Sweet home-diddly-dum-doodly. I fell in love with it the first time I saw it, as it manages to balance a touching story (which is rare in so far as it affects all the family in OFF, and doesn't just focus on the feelings of one or two individuals) with many hilarious one liners (particulally from the Flanders).

That would have been my number two on the list, maybe as high as number one - but I promised not to make a larger list.  ;)  Actually, both episodes are quite similar, showing a Lisa breaking out of her typical role. I like that a lot.  :)

I love Summer of 4'2".  ;D It's got a great story, good laughs, everyone's in character...

Apart from the ones already mentioned, here are my favorite lines...
"Okay. Calm down. It's just a bird thing. You don't control the birds. You will someday, but not now."

MARGE: Remember, there's no need to bump. Just enjoy the ride

MILHOUSE: *giggling* I signed [your yearbook] too!
LISA: Uh huh... [reads: "See you in the car! Best wishes, Milhouse"]

MARGE: Kids, behave! Don't make me get the carnie!

LISA: I know a place that's practically deserted!
[All pause.]
LISA [with "valley girl" accent]: Like, you know, whatever!
KIDS: [agreeing]

I also like Last Exit to Springfield. There's a lot of funny stuff, but the ones that stick out in my memory are mostly visual... like Santa's Little Helper yelping at Lisa's braces, Burns's memory of the "atom smashing" plant, etc. One quote I do remember:

DR. WOLFE: Maggie's teeth are coming in crooked. Has she been sucking on a pacifier?
MARGE: Mmm, not to my knowlege...
DR. WOLFE: LIAR!!

Cape Feare hands down. Never has another episode had me in so many stitches. Pretty much every episode from Seasons 3 to 5 would be in my top bracket though.

I think it's Lisa's First Word. Sweet combination of comedy and drama, and the moments with newborn Lisa are just pure cuteness. :) But Summer of 4'2" can be, too, as well as Bart vs. Thanksgiving, as well as Lisa's Wedding and many others... guys, I'm tearing.

Very hard to choose.  A lot of stuff from Seasons 3 through 6 certainly qualifies.  'Last Exit To Springfield' is a classic, 'I Married Marge' is both sweet and funny, 'Grade School Confidential' has some great sweet romance in it, and 'Marge VS. The Monorail' has a lot going for it.
